Specificity
|
This antibody was raised against the human 67LR N-terminal sequence SGALDVLQ which represents amino acids 2-9 precursor (37LRP or 'P40') (Castronovo et al.). This antibody recognises both denatured and native 67LR. Higher (110 kDa) and lower (50 kDa) bands are also detected in immunoprecipitates. Such bands have been noted before (Clement et al., Landowski et al.). The N-terminal ab2508 epitope appears to be intracellular, as non-permeablised cells do not stain with this antibody (Castronovo et al.).

Laminin, one of the major glycoproteins of basement membrane, displays multiple biological activities which are mediated through its interaction with specific cell membrane receptors. This receptor has been variously called 67 kD laminin receptor, 37 kD laminin receptor precursor (37LRP) and p40 ribosome-associated protein. Overexpression of Laminin Receptor is reportedly an independent prognostic factor in breast cancer and correlates with the dissemination of the tumor cells in bone marrow. Laminin Receptors appears to play an important role in tumor invasion and metastasis.

Notes
|
We would advise researchers use Tris (2-carboxyethyl) phosphine hydrochloride ("TCEP", available from Sigma product number 93284) as a reducing agent at a final concentration of 5mM in the treatment buffer. We believe this should produce much cleaner results than using DTT or mercaptoethanol with this antibody. Please see ab711 for further explanation.

Immunocytochemistry/ Immunofluorescence - 67kDa Laminin Receptor antibody (ab2508)
ICC/IF image of ab2508 stained human HEK 293 cells. The cells were methanol fixed (5 min), permabilised in 0.1% PBS-Tween (20 min) and incubated with the antibody (ab2508, 1:1000 dilution) for 1h at room temperature. 1%BSA / 10% normal goat serum / 0.3M glycine was used to block non-specific protein-protein interactions. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red). DAPI was used to stain the cell nuclei (blue). This antibody also gave a positive IF result in HeLa, HepG2 and MCF7 cells.

The cells were 4% PFA fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab2508, 1/1000 dilution) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
