  • SpecificityThis antibody reacts with the 68 kDa component of the polypeptide subunit. It stains neurons, neuronal processes and peripheral nerves. This antibody reacts with some neuroblastomas and gangliomas but not with other non-neuronal tumors.

    Some labs have reported an additional 200 kDa band under certain conditions. We cannot confirm if this is in fact a multimer of the 68 kDa protein.

    BALB/C mice were injected with purified neurofilaments from human brain.

  • General notesIn originator's published literature (e.g. PMID 6206727), this clone 2F11 has been shown specific to 68kDa protein however our testing on two different occasions show a 200 kDa protein band in western blot, under reduced and denaturing conditions. The 200 kDa could be either the glycosylated form of the 68kDa or 200kD a protein however we haven't done any further test to confirm it. We will welcome any feedback from researchers have who used the same clone.

  • Application notesThis antibody has been pretitered and quality controlled to work on formalin-fixed paraffin-embedded as well as acetone fixed cryostat tissue sections.
    No further titration is required.
    We suggest an incubation period of 30-60 minutes at room temperature.
    However, depending upon the fixation conditions and the staining system employed, optimal incubation should be determined by the user.

    • Involvement in diseaseDefects in NEFL are the cause of Charcot-Marie-Tooth disease type 1F (CMT1F) [MIM:607734]. CMT1F is a form of Charcot-Marie-Tooth disease, the most common inherited disorder of the peripheral nervous system. Charcot-Marie-Tooth disease is classified in two main groups on the basis of electrophysiologic properties and histopathology: primary peripheral demyelinating neuropathy or CMT1, and primary peripheral axonal neuropathy or CMT2. Neuropathies of the CMT1 group are characterized by severely reduced nerve conduction velocities (less than 38 m/sec), segmental demyelination and remyelination with onion bulb formations on nerve biopsy, slowly progressive distal muscle atrophy and weakness, absent deep tendon reflexes, and hollow feet. CMT1F is characterized by onset in infancy or childhood (range 1 to 13 years).
      Defects in NEFL are the cause of Charcot-Marie-Tooth disease type 2E (CMT2E) [MIM:607684]. CMT2E is an autosomal dominant form of Charcot-Marie-Tooth disease type 2. Neuropathies of the CMT2 group are characterized by signs of axonal regeneration in the absence of obvious myelin alterations, normal or slightly reduced nerve conduction velocities, and progressive distal muscle weakness and atrophy.

    Anti-68kDa Neurofilament antibody [2F11], prediluted images

    • ab910 staining 200kDa + 68kDa Neurofilament in human skin tissue by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections).
      Tissue was fixed in paraformaldehyde and a heat mediated antigen retrieval step was performed using citrate buffer pH 6.0. Samples were then permeabilized using 0.1% Saponin /PBS, blocked with 4% serum for 30 minutes at 25°C and then incubated with ab910 for 14 hours at 4°C. The secondary used was an Alexa-Fluor 488 conjugated goat anti-mouse polyclonal used at a 1/100 dilution.
