ABCB4 is a membrane-associated protein and member of the superfamily of ATP-binding cassette (ABC) transporters. ABC proteins transport various molecules across extra- and intra-cellular membranes. The gene encoding this protein is a member of the MDR/TAP subfamily of genes. Members of the MDR/TAP subfamily are involved in multidrug resistance as well as antigen presentation. ABCB4 is a member of the p-glycoprotein family of membrane proteins with phosphatidylcholine as its substrate. ABCB4 is the energy-dependent efflux pump responsible for decreased drug accumulation in multidrug-resistant cells, though it is not capable of conferring drug resistance. This protein mediates the translocation of phosphatidylcholine across the canalicular membrane of the hepatocyte. Defects in ABCB4 are the cause of progressive familial intrahepatic cholestasis type III (PFIC3). PFIC3 is an autosomal recessive liver disorder presenting with early onset cholestasis that progresses to cirrhosis and liver failure before adulthood. It is characterized by elevated serum gamma-glutamyltransferase levels.

Predicted band size : 141 kDa Observed band size : 170 kDa (why is the actual band size different from the predicted?) Additional bands at : 40& 43 kDa,47 kDa,75 kDa. We are unsure as to the identity of these extra bands.

Formalin-fixed, paraffin-embedded human hepatocarcinoma tissue reacted with ab71792 at a 1:50 dilution, which was peroxidase-conjugated to the secondary antibody, followed by AEC staining.
