ICC/IF: Use at a concentration of 1 µg/ml.
IHC-P: 1/300. Perform heat mediated antigen retrieval before commencing with IHC staining protocol.
WB: Use at a concentration of 1 µg/ml. Detects a band of approximately 70 kDa (predicted molecular weight: 67 kDa). We also observe a doublet band at ~100 kDa. We are unsure as to the identity of this doublet.
Not yet tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.
Antagonizes the binding of 2-5A (5'-phosphorylated 2',5'-linked oligoadenylates) by RNase L through direct interaction with RNase L and therefore inhibits its endoribonuclease activity. May play a central role in the regulation of mRNA turnover. Antagonizes the anti-viral effect of the interferon-regulated 2-5A/RNase L pathway. May act as a chaperone for post-translational events during HIV-1 capsid assembly.

Immunocytochemistry/ Immunofluorescence - ABCE1 antibody (ab32270)

ICC/IF image of ab32270 stained human HeLa cells. The cells were methanol fixed (5 min), permabilised in TBS-T (20 min) and incubated with the antibody (ab32270, 1µg/ml) for 1h at room temperature. 1%BSA / 10% normal serum / 0.3M glycine was used to quench autofluorescence and block non-specific protein-protein interactions. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red). DAPI was used to stain the cell nuclei (blue).
