Application notes

WB: 1/1000 when using colorimetric substrates such as BCIP/NBT and 1/5000 for chemiluminescent substrates. Predicted molecular weight for the full length ADAMTS7 sequence is 184 kDa, but glycosylation and the abundance of cysteine residues gives ADAMTS7 a greater apparent molecular weight of around 300 kDa on reduced SDS PAGE gels. EDTA/EGTA treatment of tissues or lysates is required to see latent zymogen. Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Relevance

ADAMTS proteases are secreted enzymes containing a prometalloprotease domain of the reprolysin type. The ADAMTS proteases function in processing of procollagens and von Willebrand factor as well as catabolism of aggrecan, versican and brevican. They have been demonstrated to have important roles in connective tissue organization, coagulation, inflammation, arthritis, angiogenesis and cell migration. A member of the metalloproteinase family containing disintegrin like domains (ADAMs), the function of ADAMTS7 is still poorly understood. ADAMTS7 most resembles ADAMTS12 in overall structure. ADAMTS7 does not have a transmembrane domain, unlike many of the ADAMs proteases, and is a secreted protein.
