Anti-ADFP antibody (ab37516) at 1/2000 dilution + E. coli-derived fusion protein at 2.5 µg
Secondary
Goat anti-IgY-HRP at 1/1000 dilution
Predicted band size : 48 kDa
Observed band size : 75 kDa (why is the actual band size different from the predicted?)
A fusion protein was used in this western blot; hence the difference in molecular weight compared to native protein.
ab37516 staining ADFP in Human THP-1 cells by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with formaldehyde, permeabilzed with 0.05% saponin and blocked with 1% BSA for 2 hours at 22ºC. Samples were incubated with primary antibody (1/400 in 1% BSA, PBST) for 16 hours at 4ºC. An Alexa Fluor®568-conjugated goat anti-chicken monoclonal (1/500) was used as the secondary antibody. ADFP was localized on the periphery of small lipid droplets around the cell.
