Our Abpromise guarantee covers the use of ab49139 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
WB: Use a concentration of 0.5 µg/mlPredicted molecular weight: 69 kDa.
IHC-P: Use a concentration of 5 µg/mlPerform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.
Vitamin E binding protein. May transport vitamin E in body fluids under conditions where the lipoprotein system is not sufficient or across the blood-brain barrier.
Belongs to the ALB/AFP/VDB family.
Contains 3 albumin domains.
N-glycosylated.
Secreted.

All lanes : Anti-AFM antibody (ab49139) at 0.5 µg/ml
Lane 1 : Molecular weight marker
Lane 2 : HepG2 cell lysate at 10 µg
Secondary
HRP conjugated anti-Rabbit IgG at 1/50,000
developed using the ECL technique
Predicted band size : 69 kDa
Observed band size : 69 kDa
Gel concentration: 12%
Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - AFM antibody (ab49139)

IHC image of ab49139 staining in human normal liver formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab49139, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
