ordered and received the products ab12104 and ab16555. I have already tried them with good results for the first and bad results for the second. As you can see fron the attached pictures, anti Aph-1 works as described and recognizes a band of the correct MW plu other bands which are described as possible aggregates or proteolysis fragments. As for anti Pen2, the number of aspecific bands with very strong signal is incredibly high; there is also a band of about 12 kd which could be the correct one, but it is very difficult to be sure of the identity of this band. I understand that it is a whole serum, still the n° of aspecific bands is way too high! Do you have the antigen peptide in order to see which of the bands disappears? Can you send an aliquot to me? Since your web page I read: "Quality is important to Abcam. If PEN2 antibody (ab16555) does not perform as described on this datasheet, notify us within 90 days of delivery using this form, so that we can help you or offer you a replacement or a refund" I have put on hold the payment of the products, but maybe you can tell me what else I have to do. I am obviously repeating my western with different conditions, but I am afraid that without a positive and negative control (or the antigen peptide) it will be difficult to get great improvement. We have received the following information from the source of ab16555: This protein is an integral membrane protein (Potential). It is predominantly located in the endoplasmic reticulum and in the cis-Golgi. Tissue specificity: Widely expressed. Expressed in leukocytes, lung,placenta, small intestine, liver, kidney, spleen thymus, skeletal muscle, heart and brain. Please refer to the following journal article, as it is specific for this particular antibody in both WB and IP on mouse neuroblastoma cells. It also provides images. Luo, W, et al. Expected molecular weight was 30kDa. The band observed between 20-25kDa is likely to be an isoform of Aph1a.
We are unsure as to the identity of the minor band seen at 75kDa.

ab12104 staining APH1a in mouse brain tissue section (cortex) by Immunohistochemistry (PFA perfusion fixed frozen sections). Tissue from 4% PFA perfused animals underwent overnight fixation in 4% paraformaldehyde, cryoprotected in 30% sucrose and cut using cryostat.The primary antibody was diluted, 1/1000 (PBS + 0.3% Triton X100) and incubated with sample for 18 hours at 20°C. An abcam antibody ab60314, Chromeo488 conjugated goat polyclonal to rabbit IgG, diluted 1/1000 was used as secondary. ab12104 produces a very pretty staining in neuronal processes in the cortex and the hippocampus.
