Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - APLP2 antibody (ab58583)

Immunohistochemistical detection of APLP2 using antibody ab58583 on (Formaldehyde-fixed paraffin-embedded rat lumbar DRG sections). Antigen retrieval step: heat mediated; Buffer used: Citric acid pH6; Blocking step: 1% BSA for 10 mins@ rt°C. Primary antibody ab58583 diluted @ 1/1000, incubated for 16 hours in TBS/BSA/azide. Secondary antibody: anti Rabbit IgG conjugated to biotin (1/200). The immunostaining appears to be localised to the cytoplasm: a widely distributed punctate positivity is seen in DRG cells. Note that DRG nuclei are "absent": Citric acid HIER pH6, in my hands, tends to hydrolyse DNA in highly metabolically active cells. NB: other nuclei are very well stained. I overstained this section with Hx: note the blue staining of the myelin sheaths.

Antigen retrieval step: Heat mediated in citric acid pH6 buffer. Blocking step: 1% BSA for 10 mins @ RT°C. Primary antibody ab58583 used at 1/2500 incubated for 2 hours in TBS/BSA/azide. Secondary antibody: anti Rabbit IgGs conjugated to biotin (1/200). When I probed this Ab on TASTPM brain I found a very high elevation of positivity in amyloid plaques (the same in Tg2576 mouse brain). The lower image is included to confirm that APLP2 is demonstrated in amyloid plaques: both images were taken using the same capture conditions.
