Does item#ab2905 detect phosphorylated ATR?

ANSWER:

 

Thank you for your enquiry.

This antibody is not phospho-specific, but since it a polyclonal raised to a fusion protein using 60 amino acids of ATR, I would expect the antibody to have multiple epitopes and detect total ATR, including phosphorylated protein. However, I don't see that this has been specifically tested.

DESCRIPTION OF THE PROBLEM Rather than a problem, we are concerned about the complete transfer of this huge ATR protein (see below). It would be great if you could tell us the transfer conditions you used to get the nice WB pictures you show for ab4471 and ab2905: immersion or semi-dry, buffer composition, current, time.

We are running 6% SDS-PAGE (we cannot afford comercial gradient gels).

We have received the following information regarding ab2905:

It is best to try a high amp for about an hour. We recommend trying at 1 amp for 1 hour using appropriate methods to keep the system cool. We used 3 ml of 5% nonfat dried milk in TBST. Primary Ab. dilution of 1:1000 for 1 hour at room temp. Washed 3X with TBST buffer for 8 min at room temp. Each gently agitating solution. Secondary Ab 1:15000 in blocking buffer for 1 hour at room temp. Wash 3X with TBST for 8 min with agitation. Exposed for 15 seconds by ECL detection.

I would like to know if the rabbit polyclonal antisera against ATR (ab2905) cros-reacts with ATM, and if it has been used for ATR immunocomplex kinase assays.

ANSWER:

 

It has not been tested for its reactivity with ATM nor has the antibody been used in kinase assays, therefore we have no further information on these.
