    The customer kindly phoned to ask about differences between these two products. They are working with duodenum tissues which are very active in iron absorption and are wondering if this high iron contenct will affect the activity assay as well.

    I have been in touch with the labs regarding use of ab109712 &ab83459 for duodenumtissue because of its iron aborbence.

    In regards to ab109712:Although we have not tested in this tissue type, there have been studies on aconitase and duodenal iron – one paper shows no effect.http://www.ncbi.nlm.nih.gov/pubmed/8541321

    In other studies:http://www.ncbi.nlm.nih.gov/pubmed?term=iron%20dudenum%20aconitase Ppresumably the iron is sequestered after absorption preventing significant damage.



    In regards to ab83459:We do not think this will be a problem, but we have not tested duodenum.

    There are two kits in our catalogue targeting Aconitase: ab83459 and ab109712. Both are compatible with frozen tissues, as long as they are frozen appropriately preserving the enzymatic activity.

    I want to detect the activity of bacteria's Aconitase, whether your product Aconitase Assay Kit suit the bacteria?

    As far as we are aware, this kit has never been tested with samples from bacteria. It might be possible, as the assay allows detection of c-aconitase and m-aconitase (from mitochondria), however this can not be guaranteed as it has not been tested. Maybe Walden et al (From bacteria to mitochondria: Aconitase yields surprises) can be of help.

    cysteine and iron-sulfur protein amount/concentration? want to use own lysis buffer instead of assay buffer?

    For 1X Cell lysis buffer: - 20mM Tris-Hcl - 150mM NaCl - 1mM Na2EDTA - 1mM EGTA - 1% Triton - 2.5mM sodium pyrophosphate - 1mM beta-glycerophosphate - 1mM Na3VO4 - 1ug/ml leupeptin

    ANSWER:

     

    The assay buffer is Tris based with some nonionic detergent. We are unable to be more specific. Other components included in your lysis buffer have not been tested for compatibility with this kit.
