Assay conditions: The 50 µl reaction mixture contained 50 mM Tris-HCl, pH 8.0, 12.5 mM MgCl2, 0.1 mM nicotinamide, 0.4 mM PRPP, 2 mM ATP, 30 µg/ml of alcohol dehydrogenase, 10 µg/mL of NMNAT, 1.5% alcohol, 1 mM DTT, 0.02% BSA. Reactions were incubated at room temperature for 30 min, and the increase in NADH fluorescence (Ex 340 nm/Em 460 nm) was measured. -

Catalyzes the condensation of nicotinamide with 5-phosphoribosyl-1-pyrophosphate to yield nicotinamide mononucleotide, an intermediate in the biosynthesis of NAD. It is the rate limiting component in the mammalian NAD biosynthesis pathway. -
