Question 1

Thursday 17-August-2006

Dear Colleague, I hope you can help me with this enquiry. Regarding ab10592 (Activin Receptor Type IA antibody), could you please provide the immunogenic recombinant fragment sequence? Any additional information would help the customer. Thanks! Have a good day. Regards, Cindy

ANSWER:

Dear Dr. Fiori,

Thank you for your enquiry.

Regarding ab10592 (Activin Receptor Type IA antibody), this antibody was raised against the extracellular domain of rhActivin RIA (ALK-2, protein accession number, NP_001096.1), corresponding to Met1-Val124. Specifically, a DNA sequence encoding the extracellular domain of the human Activin RIA protein sequence (Dijke, P. et al., 1993, Oncogene 8:2879 - 2887) was fused to the carboxy-terminal Fc region of human IgG1 via a polypeptide linker. The chimeric protein was expressed in a mouse myeloma cell line, NS0. Please note that this antibody cannot be used for detecting the other domains (e.g. cytoplasmic, protein kinase, ATP, etc).

I hope this information helps.

Question 2

Wednesday 22-March-2006

As far as I know, P36894 is Alk3. I agree the nomenclature is confusing, but let's faith it, it wouldn't be the first time. The Swiss prot reference is to a 60 kDa, 532 aa acid peptide with the appropriated domains. So I have been assuming they are one and the same. There are a number o splice variants which are associated with a fairly rare condition called Juvenile polyposis. So it may be worth checking exactly where the peptide sequence you have used to raise the ab is located in the protein.

Personally I'm more interested in its function in human renal epithelial cells and was hoping to use it in Western Blotting but would dearly love to try and use it in immuno-precipitation. Why do you guys never test these interesting antibodies for IP.

So what discount will you give me on this product then ? we'll review it for you.

ANSWER:

Thank you for getting back to me and for your insight into this antibody target. I have updated the SwissProt links of this antibody with immediate effect.

I can offer you a 15% discount on the antibody in return for feedback on how the antibody behaves by immunoprecipitation (it has already been shown to work by western blotting). I would ask that you provide us with details of how it performed by navigating to the datasheet and clicking on the "Abreviews" tab.

If you are interested in this offer please e-mail me and I will provide you with a unique discount code that can be used during the ordering process.

Question 3

Monday 20-March-2006

I was interested in buying this produc, which you describe as being an Alk3 antibody, however both the OMIM and SwissProt links you give are for Alk2.

Could you please calrify which Alk this antibody recognises

Thank you.

ANSWER:

Thank you for your enquiry. Please let me apolagise for the confusion. This is an antibody against Alk3/Activin Receptor Type IA. The only SwissProt reference that I could find against this target is BMR1A_HUMAN (P36894). However, this is bone morphogenetic protein receptor type IA [Precursor] and I am not certain that this corresponds to this target. It may be that the protein has not been characterized in SwissProt. I would appreciate some guidance if you know different.

I look forward to hearing from you.
