    Adiponectin, also known as Adipocyte Complement-Related Protein of 30kDa (ACRP30), is a secreted serum protein expressed exclusively in differentiated adipocytes. Studies indicated that decreased plasma adiponectin concentration is associated with obesity, insulin resistance, essential hypertension, inflammation and atherosclerosis, and acute myocardial infarction. On the other hand, increased adiponectin level leads to nephrotic syndrome.

    ab108784 Adiponectin Rat ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the detection of rat Adiponectin in urine, plasma, serum, and cell culture supernatants. This assay employs a quantitative sandwich enzyme immunoassay technique that measures rat Adiponectin in less than 3 hours. A polyclonal antibody specific for rat Adiponectin has been pre-coated onto a 96-well microplate with removable strips. Adiponectin in standards and samples is sandwiched by the immobilized polyclonal antibody and biotinylated polyclonal antibody specific for rat Adiponectin, which is recognized by a streptavidin-peroxidase conjugate. All unbound material is then washed away and a peroxidase enzyme substrate is added. The colour development is stopped and the intensity of the colour is measured.

  • FunctionReceptor for globular and full-length adiponectin (APM1), an essential hormone secreted by adipocytes that acts as an antidiabetic. Probably involved in metabolic pathways that regulate lipid metabolism such as fatty acid oxidation. Mediates increased AMPK, PPARA ligand activity, fatty acid oxidation and glucose uptake by adiponectin. Has some high-affinity receptor for globular adiponectin but low-affinity receptor for full-length adiponectin.
  • Tissue specificityWidely expressed. Highly expressed in skeletal muscle. Expressed at intermediate level in brain, heart, spleen, kidney, liver, placenta, lung and peripheral blood leukocytes. Weakly expressed in colon, thymus and small intestine.

  • DomainThe N-terminus is known to be cytoplasmic while the C-terminus is known to be extracellular.

    I have diluted the standard which is labeled as 1x1.6ug in 4mls diluent to produce the recommended concentration of 400ng/ml. However the standard curve essentially is all blank. there is no color change even at the 100ng standard concentration.

    Standard in this kit was 1.6 ug/bottle. The customer should add 4 ml to get standard solution of 400 ng/ml and then do 1:4 dilutions for the first standard point to get working solution of 100 ng/ml
