This isozyme may play a role in skeletal mineralization.
Defects in ALPL are a cause of hypophosphatasia (HOPS) [MIM:146300]. HOPS is an inherited metabolic bone disease characterized by defective skeletal mineralization. Four hypophosphatasia forms are distinguished, depending on the age of onset: perinatal, infantile, childhood and adult type. The perinatal form is the most severe and is almost always fatal. Patients with only premature loss of deciduous teeth, but with no bone disease are regarded as having odontohypophosphatasia (odonto).
Defects in ALPL are a cause of hypophosphatasia childhood type (HOPSC) [MIM:241510].
Defects in ALPL are a cause of hypophosphatasia infantile type (HOPSI) [MIM:241500].

Undifferentiated human embryonic stem cells (H9 line) were stained with the tissue non-specific alkaline phosphatase antibody, ab17973. The cells were positive for alkaline phosphatase with staining localised on the surface membrane.
Dilution used: 1/50
Note that the feeder cells in the bottom left hand corner of the plate are not stained by this antibody.

ab17973 staining Alkaline Phosphatase, Tissue Non-Specific in human adrenal gland tissue by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded tissue sections). The sections were fixed in paraformaldehye prior to blocking with 10% serum for 1 hour at 20°C. The primary antibody was diluted 1/500 and incubated with the sample for 12 hours at 4°C. An HRP-conjugated rabbit anti-mouse polyclonal was used as the secondary antibody, diluted 1/200.
