Thank you for your reply. Before I give all the details that you requested I just wanted to say that abcam is my first choice in terms of antibodies and I have not been disappointed so far.

1. The purchase order is: ######

2. Tissue type: Rat kidney cortex

3. Conditions: I have tried a range – 1to1000, 1to2000, 1to4000, 1to8000 dilutions, and I obtained linearity with 1to2000 dilution and 40ug, 60ug and 80ug of protein. I am running western with 60ug of protein and 1to2000 dilution of primary antibody. The incubation with primary antibody was indeed carried out overnight in 4oC.

4. Storage: On the delivery day I spin down (in 4oC) and aliquoted antibody to 5ul. Eppendorf tubes were additionally wrapt with paraffin film to prevent any losses. Tubes were kept at -20oC.

I would be grateful for another tube of antibody as I have still got more samples to run. If you need any additional information please let me know.

Dear Madam/Sir,

I have been using your ab19134 anti- AT2R (rabbit polyclonal primary antibody) for quite a long time. It worked beautifully until last week.

I ordered from you in the end of last year 2 tubes (2x50ug) of ab19134, both lot: GR-55294-1. The antibody was aliquoted to the required amount and stored as advised. Until recently I have been using aliquots from one tube and it was fine. This week I used antibody aliquots from the second tube and my western did not work properly. Western blot: I am using Amersham fluorescent double staining. On the same membrane, at the same time I scan for bands of AT2R and Histone. As you can see on the membrane scanned on 06-03-12 histone is fine (Please see attached images). The membrane from 01-03-12 is the good one but on the 06-03-12 I can barely see the band for AT2R. I did not change anything but the primary antibody. Since I treated both of the tubes in the same way from the arrival I suspect that the problem occurred before ab19134 was shipped. Therefore, would it be possible to get a refund or another 50ug of ab19134?
