  • Product overview

Human Apolipoprotein AI (Apo A1) comprises about 70% of the high-density lipoproteins (HDL) protein mass and Apo A1I another 15–20%. Apo A1, a 243-amino acid molecule that contains a series of highly homologous amphipathic alpha-helices, is a 28-kDa single polypeptide that lacks glycosylation or disulfide linkages. About 5–10% of human plasma Apo A1 exists in a lipoprotein-unassociated state. Apo A1 appears to have effects on the atherosclerosis inhibition, reverse cholesterol transport and anti-inflammation. Oxidation of specific amino acid residues in Apo A1 may contribute to atherogenesis by impairing cholesterol efflux from macrophages. A majority of HDL functionality is derived from the ability of Apo A1 to sequester phospholipid and cholesterol and interact with plasma enzymes and cellular receptors. During reverse cholesterol transport, HDL interacts with lecithin: cholesteryl acyltransferase (LCAT) and cellular receptors, including ATP-binding cassette transporter protein A1 (ABCA1) and the scavenger receptor class B type I in an ordered fashion that is reflected by HDL particle lipid composition. A high-affinity HDL receptor for Apo A1 is beta-chain of ATP synthase on the surface of hepatocytes. The plasma concentration of Apo A1 is one of the best indicators of susceptibility to cardiovascular disease.

Ab108804 Human Apo A1 ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for detection of human Apo A1 in plasma and serum. This assay employs a quantitative competitive enzyme immunoassay technique that measures human Apo A1 in less than 3 hours. A polyclonal antibody specific for human ApoA-1 has been pre-coated onto a 96-well microplate with removable strips. Apo A1 in standards and samples is competed with a biotinylated Apo A1 sandwiched by the immobilized antibody and streptavidin-peroxidase conjugate. All unbound material is then washed away and a peroxidase enzyme substrate is added. The color development is stopped and the intensity of the color is measured.

Below is the information for the standard specific to your lot:

I contacted the laboratory, and they responded that the Apo A-1 standard should be in micrograms, not nanograms. The current lot's standard is delivered at 60ug and 3mL of diluent would make the 20ug/ml solution. If you respond with your specific lot...
