- Involvement in diseaseDefects in AQP2 are the cause of diabetes insipidus nephrogenic autosomal (ANDI) [MIM:125800]; also known as diabetes insipidus nephrogenic type 2. ANDI is caused by the inability of the renal collecting ducts to absorb water in response to arginine vasopressin. It is characterized by excessive water drinking (polydypsia), excessive urine excretion (polyuria), persistent hypotonic urine, and hypokalemia. Inheritance can be autosomal dominant or recessive.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Aquaporin 2 antibody - C-terminal (ab85876)This image is courtesy of an Abreview submitted by Ross Benikab85876 staining Aquaporin 2 - C-terminal in Rat kidney tissue sections by Immunohistochemistry (IHC-P - paraffin-embedded sections). Tissue was fixed with 10% NBF and blocked with 25% serum free protein block for 5 minutes at 25°C; antigen retrieval was by heat mediation with a Tris/EDTA (pH 9.0) buffer. Samples were incubated with primary antibody (1/5000 in serum free antibody diluent) for 30 minutes. An undiluted HRP-conjugated Goat anti-rabbit IgG polyclonal was used as the secondary antibody. -
ICC/IF image of ab85876 stained HepG2 cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab85876, neat) overnight at +4°C. The secondary antibody (green) was ab96899, DyLight® 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
