Relevance

The budding yeast INO80 complex is a conserved ATP-dependent nucleosome remodeler containing actin-related proteins Arp5 and Arp8. Strains lacking INO80, ARP5, or ARP8 have defects in transcription. Arp8 is an actin related protein that is probably involved in transcription regulation via its interaction with the INO80 complex, a chromatin remodeling complex.

ab12098 recognises a band of approximately 100kDa corresponding to Arp8 using purified INO80 complex. The antibody does not recognize Arp8 using S. pombe or S cerevisiae whole cell extracts but it is possible that this could be improved using different blocking conditions.

Predicted band size : 100 kDa Observed band size : 100 kDa Additional bands at : 95 kDa (possible degradation product).
S. pombe FLAG-tagged-INO80 complex was immunoprecipitated (lane2) and immunoblotted using anti-Arp8 antibodies (ab12098).
Arp8 a member of the INO80 complex was detected as at approximately 100 kDa, which is predicted to be the higher band of the doublet. The lower band of the doublet may represent a proteolytic cleavage product.
