Application notes

IHC-P: 1/50. Perform heat mediated antigen retrieval via the pressure cooker method before commencing with IHC staining protocol. Use streptavidin-biotin amplification (see recommended protocol).
Flow Cyt: Use at a concentration of 1 µg/ml.
Incubate cells on ice for 30 min in 50 µl FACS buffer (PBS, 5% Fetal calf serum, 0.02% azide) containing 1µg/ml of Buffy-2-FITC antibody (testing conditions for HEK 293T cells (5 x 105) mock transfected or transfected with an expression plasmid enabling surface expression of full length hBAFF).

Not tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.

Function

Cytokine that binds to TNFRSF13B/TACI and TNFRSF17/BCMA. TNFSF13/APRIL binds to the same 2 receptors. Together, they form a 2 ligands -2 receptors pathway involved in the stimulation of B-and T-cell function and the regulation of humoral immunity. A third B-cell specific BAFF-receptor (BAFFR/BR3) promotes the survival of mature B-cells and the B-cell response.

Tissue specificity

Abundantly expressed in peripheral blood Leukocytes and is specifically expressed in monocytes and macrophages. Also found in the spleen, lymph node, bone marrow, T-cells and dendritic cells. A lower expression seen in placenta, heart, lung, fetal liver, thymus, and pancreas.

Staining of paraffin section of synovial tissue from a patient with Rheumatoid Arthritis. Arrows  indicate vessels staining positive for BAFF using ab16082.
Method: Tissues are in paraffin sections. Sections are de-waxed and rehydrated, then cooked. Sections are stained in wet boxes, rehydrated and finally prepared as coverslip slides using Eukitt solution.

Ab16082 detects membrane-bound human BAFF by FACS.
Method: HEK 293T cells (5 x 10e5) were mock transfected (thin line) or transfected with an expression plasmid enabling surface expression of full length hBAFF (thick line). Cells were incubated on ice for 30min in 50μl FACS buffer (PBS, 5% fetal calf serum, 0.02% azide) containing 1µg/ml of ab16082. After washing, cells were analyzed by flow cytometry.
