Question 2

Tuesday 21-February-2012

Hello Abcam team, I would like to do IHC in spinal cord for BDNF and Cholera toxin B. Can I perform double fluorescent staining simultaneously according to your protocol?. What is the best antibody for Spinal cord tissue, according to your experience, and what is the best dilution?. 1/500 versus 1/100. Thanks

ANSWER:

Thank you for your enquiry.

For BDNF, I can suggest choosing the antibody that has been tested in IHC-P (paraffin embedded sections) or IHC-Fr (frozen sections). Whichever one of these applications you are using, I recommend to select an antibody that has been tested. This will ensure it is covered by our guarantee. I have provided a link below to the search results from our website for BDNF antibodies tested in IHC-P and IHC-Fr. I can recommend to review the online datasheets for further information (including suggested dilutions to use in IHC) and choose one that is also tested in the species you are using:

http://www.abcam.com/index.html?pageconfig=searchresults&search=BDNF&pt=1&sk=app&sv=78&sn=IHC-Fr&l=3&fViewMore=1

Regarding Cholera B toxin antibodies, we have several antibodies to beta subunit cholera toxin. The following is tested in IHC-Fr:

ab62429 Anti-beta subunit Cholera Toxin antibody [23043] Mouse monoclonal
http://www.abcam.com/index.html?datasheet=62429 http://www.abcam.com/index.html?datasheet=62429.

If you wish to use this in double staining with the BDNF antibody, you will need to select a BDNF antibody that is not raised in mouse. For example, you could try a rabbit polyclonal. This is to prevent the secondary antibodies from crossreacting with both primary antibodies. The protocol for double staining would need to be individually optimized in your own laboratory as these antibodies have not been tested together yet.

Please note that ab62429 has not been tested in IHC-P. If you would like to try this, please do not hesitate to let me know before you purchase as you may be eligible for our testing discount program for this.

I hope this will be helpful to you. If you have any further questions, please do not hesitate to ask.

Question 3

Monday 05-December-2011

To whom it may concern,     I would like to choose a product for the BDNF Elisa. Currently our kit needs an anti-BDNF mAb for coating wells (NUNC maxisorp). I am not sure which antibody I should buy, the ab108319 or the ab46176? Could you please give me some suggestion? I work with the cell line neuro-2a. Thanks.   Best,

ANSWER:

Thank you for contacting us and for your interest in our products.

I think ab108319 (www.abcam.com/ab108319) or ab108383 (www.abcam.com/ab108383) would be suitable since you are looking for a monoclonal antibody. However, please note that none of these 2 antibodies is purified. Furthermore, as far as we are aware, ab108319 and ab108383 have never been tested in ELISA. All tested applications are specified on our datasheets, and these are updated as soon as any new information is brought to our attention.

If you would like to test one of these antibodies in ELISA, please contact our Scientific Support team prior to the purchase by replying to this message as you may be eligible for our testing discount program.

Otherwise, we like to encourage all of our customers to submit an Abreview via the online product datasheet. We always appreciate customer feedback, whether positive or negative, and we make all product information available to researchers. Plus, each Abreview earns Abpoints that can be used for discounts on future purchases or rewards such as Amazon.com gift certificates.

To find out more about our Abreview system, please see the following link:

I hope this information is helpful. Please do not hesitate to contact me for any further advice or information in this regard.

Question 5

Tuesday 22-November-2011

Dear abcam technical support team:

This customer raises an enquiry about Anti-BDNF antibody. This customer would like to conduct sandwich ELISA assay with mouse balf and serum sample.

Therefore, he wants to ask for your help to offer some suitable selection which can repair each other and work well.

Could you please help this customer to solve the problem?

Thanks for your kindly assistance.

Best regards

ANSWER:

Thank you for contacting us. Having reviewed the products in our catalogue I would suggest rabbit polyclonal Anti-BDNF ab46176 and Chicken polyclonal Anti-BDNF ab28952 may be suitable for your application. They have not, as far as we know, been used in Sandwich ELISA so far. The immunogen sequences used for each antibody is however different and should therefore both be able to recognise the protein simultaneously. However we cannot guarantee that this will work.

If you were to purchase these antibodies to try with sandwich ELISA we would be able to offer a testing discount. This would entitle you to a free primary antibody of your choice if you share the results of the experiment with us, whether positive or negative, through an abreview. More information on this offer can be found here:

www.abcam.com/collaborationdiscount

If you would be interested in this offer please let me know as the discount code needs to be issued prior to purchase.

We have several secondary antibodies which would be suitable in conjunction with these primary antibodies: For ab46176, secondary antibodies ab97080 (HRP conjugated) or ab98506 (Biotin conjugated) could be used. Or if using ab97142 as the detection antibody the secondary antibodies ab97142 (Alkaline phosphotase conjugated) or ab97150 (HRP conjugated) could be used.

I hope this information has been of help. If you require any further information please do not hesitate to let us know.
