Application notes

|
Recommended dilutions IHC-P: Use at an assay dependent dilution PMID 17959825. Do not perform antigen retrieval. I-ELISA: Use at a concentration of 0.5 - 1 µg/ml to detect a limit of ~3 ng/well recombinant human BMP-2 at 50 and 100 ng/lane under non-reducing and reducing conditions, respectively. WB: Use at a concentration of 1 - 2 µg/ml. Detects a band of approximately 17-18 kDa.
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Relevance
|
BMP2 belongs to the transforming growth factor-beta (TGFB) superfamily of secreted growth factors. It is a disulfide-linked homodimer and induces bone and cartilage formation. In addition to its osteogenic activity, BMP2 plays an important role in cardiac morphogenesis and is expressed in a variety of tissues including lung, spleen, brain, liver, prostate ovary and small intestine. The functional form of BMP2 is a 26 kDa protein composed of two identical 114 amino acid polypeptide chains linked by a single disulfide bond. BMPs control fundamental events in early embryonic development, organogenesis and adult tissue homeostasis.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - BMP2 antibody [65529.111] (ab6285)
ab66285 staining BMP2 in Human small intestine tissue sections by IHC-P (formaldehyde-fixed paraffin-embedded sections). Tissue was fixed with formaldehyde and blocked with 1.5% serum for 10 minutes. Antigen retrieval was by heat mediation in citrate (pH6.0). Samples were incubated with primary antibody (1/100 in diluent buffer) for 1 hour. An undiluted HRP-conjugated Goat polyclonal to mouse IgG was used as secondary antibody This image is courtesy of an anonymous Abreview
