The antibody can recognize the monomer of rhBMP-2 (reducing condition) but poorly recognize the dimer/mature rhBMP-2 (with glycosylation). Question: what is the immunogen of this antibody? (E-coli or CHO derived human BMP-2, monomer or homodimer, with or without glycan)

ANSWER:

 

The immunogen is an E. coli-derived 26kDa homodimeric disulfide-linked human BMP-2 which is not glycosylated.

You can try non-reducing conditions (omit DTT or b-mercaptoethanol) and boiling less time in SDS loading buffer to avoid disrupting dimers.
