Thank you for your enquiry. Both ab8243 and ab24451 were generated using highly purified lethal factor antigen of Bacillus anthracis. These two antibodies can be used together in a sandwich ELISA where ab8243 is the coating antibody and ab24451 is the conjugate antibody. To use these antibodies in ELISA we would recommend start at a concentration of 5 ug/ml but the end-user should optimize based upon results. The calibration curve for ELISA on the datasheet states that ab8242 was used for coating, and ab8243 as a HRP conjugate. Is ab8243 HRP conjugated? |

your B. anthracis antibody #AB8242 should be used as the "conjugate" along with AB8243 as the coating antibody. Does this mean that AB8242 is conjugated (to HRP?)? If it is not conjugated, which secondary antibody is recommended? How can it be avoided that the secondary antibody does not only detect AB8242 but also the coated AB8243 (both are mouse monoclonals)?

Thank you for your enquiry. Our source has replied: "The normal presentation of the antibodies is non-conjugated, but we are also offering them in conjugated form. We are normally not using anti-mouse IgG secondary antibodies, we are using hrp-conjugated anti-B.anthracis antibodies (HRP-conjugated clone BAL0105, when using the pair BAL0105-BAL0106 in sandwich ELISA)".
