Customer is interested in phospho S112 and phospho S136 Bad antibodies for use in WB. Do any detect the endongenous protein?

Fo ab10428, should a total Bad antibody be used to IP with first?

ANSWER:

 

Thank you for your enquiry. It does not appear that any of the phospho S112 and phospho S136 Bad antibodies that we currently have (see below) have been used to detect the endogenous protein. The exception is ab1435 - as you can see from the WB image on the online datasheet, there is a faint band detected in untreated COS cell lysate. However, it does appear that endogenous levels of phospho-Bad are indeed very low.

Bad (phospho S112) antibodies:

Ab1435 - tested in WB with TPA treated and un-treated COS cell lysate.

Ab4770 - tested in WB with HeLa cells treated with EGF.

Ab10428 - a total BAD antibody was used to IP samples prior to probing with ab10428.

Bad (phospho S136) antibodies:

Ab4771 - tested in WB with 293 cells expressing Flag-BAD.

Ab10429 - a total BAD antibody was used to IP samples prior to probing with ab10429.
