Question 1

Monday 16-January-2012

I just have a simple question about the concentration of the antibody supplied (or total micrograms). The only info I found was the volume (500ul). I haven´t run the analysis yet - waiting for your response.
SAMPLE human embryonic stem cells PRIMARY ANTIBODY planning to use 0,5ug / 500,000 cells DETECTION METHOD flow cytometry POSITIVE AND NEGATIVE CONTROLS USED - w/o primary Ab - P3X ANTIBODY STORAGE CONDITIONS 4°C SAMPLE PREPARATION 0,5microM EDTA in PBS fofr collection 2%FBS in PBS for further handling NUMBER OF CELLS USED 500,000 PERMEABILIZATION STEP no BLOCKING CONDITIONS no SECONDARY ANTIBODY Caltag anti-mouse-FITC HAVE YOU RUN A "NO PRIMARY" CONTROL? Yes ADDITIONAL NOTES I just have a simple question about the concentration of the antibody supplied (or total micrograms). The only info I found was the volume (500ul). Thanks a lot

ANSWER:

Thank you for your inquiry.

Indeed this antibody is an Ascite. The antibody concentration is therefore not known. We do however estimate that an average ascite hasa concentrationfrom 1 to 10 mg/ml, with about 90% of specific antibodies.

We do guarantee this antibody for the use in flow cytometry. If you would like however, you can purify this antibody to obtain the Ig fraction and quantify it. As this antibody is a mouse IgG3, you can use both, protein A or protein G for its purification. Both do have however only weak bindingfor the mouse IgG3, compared to binding to other Igs(http://www.abcam.com/index.html?pageconfig=resource&rid=11385table at the bottom). We do also have such purification kits, for example ab102783. http://www.abcam.com/index.html?datasheet=102783.

I hope this information is helpful. Please do not hesitate to contact us should you have any further question.
