Our Abpromise guarantee covers the use of ab58825 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
WB: 1/1000Detects a band of approximately 42 kDa (predicted molecular weight: 35 kDa).
ELISA: Use at an assay dependent dilution.
IHC-P: Use at an assay dependent dilution.
Binds tightly to hydroxyapatite. Appears to form an integral part of the mineralized matrix. Probably important to cell-matrix interaction. Promotes Arg-Gly-Asp-dependent cell attachment.
N-glycosylated; glycans consist of sialated and core-fucosylated bi-, tri- and tetraantennary chains.
O-glycosylated at eight sites; mucin-type glycans contain Gal, GlcNAc, GalNAc and terminal NeuAc.
Sulfated on either Tyr-313 or Tyr-314.
Secreted.

Anti-Bone Sialoprotein antibody [ID1.2] (ab58825) at 1 µg/ml + MG-63 Human Osteosarcoma Whole Cell Lysate at 20 µg
Secondary
Goat polyclonal Secondary Antibody to Mouse IgG - H&L (HRP), pre-adsorbed (ab97040) at 1/5000 dilution
developed using the ECL technique
Performed under reducing conditions.
Predicted band size : 35 kDa
Observed band size : 42 kDa (why is the actual band size different from the predicted?)
Additional bands at : 36 kDa. We are unsure as to the identity of these extra bands.
Exposure time : 20 minutes
Bone Sialoprotein contains a number of potential glycosylation sites (SwissProt) which may explain its migration at a higher molecular weight than predicted.
ab58825 has not yet been referenced specifically in any publications.
