Please recommend a negative control for this antibody.

ANSWER:

 

I was informed that the best negative control is affinity purified goat IgG antibody to a protein that is not expressed on the target cells cells/tissues to be examined. For example affinity purified Goat IgG antibody to Trail-2 or Trail-3 receptor antibody will be a good negative control. In our studies these antibodies failed to stain lymphoid cells. Other similar antigen-affinity purified goat IgG antibodies can also be used as negative controls.

I have recently purchased this antibody from you based on your information sheet. It states that the antibody cross reacts with mouse CCR6. My Western was negative, so I looked into why this could be. I referred to the paper you cited as using the antibody, but was unable to find any reference to it in the paper (GPR-CY4 = CCR6). Secondly the homology between the Human immunogen and the mouse peptide sequence is less than 50%.

Your data sheet indicates that the product has been tested in mouse and Human. I would be very grateful if you could send me data regarding the western blotting of your antibody to mouse CCR6.

ANSWER:

 

The source of ab1659 has replied to me the following:

"We agree with the customer that antibody to human CCR6 should not react with the mouse CCR6 because of the significant sequence differences. However, work with mouse white bloods cells showed clearly positive blot results. At the present time we are looking at that data with more scrutiny. From now on the datasheets should be modified recommending that the antibody should be sold only for human CCR6 receptor only.

We know that the extracellular domain of CCR6 does not extend up to amino acid 46 but due to glycosylation sites at the amino terminus of the extracellular domain of human CCR6. We chose to include additional residues at the C-terminal end of the peptide due to scientific and technical reasons that are confidential."

Following this correspondence we have decided to change our datasheet to reflect your findings and those of the originator. I would like to offer you our sincere apologies for this problem and arrange a refund to be sent to you.
