I found a similar antibody from LSBio (raised against the same peptide) that says it reacts with mouse. I do not necessarily want to try ab78429 again. Could you send me ab1659 so I can see if I can make it work? Thanks for the quick response and great service! This is why we buy so much from you! Thanks again. |

ANSWER: |
I will be happy to send the ab1659 to you as a replacement for ab78429 however I did look at the LSBio antibody I think you are talking about, cat #LS-C83958 and they only predict that it will react with mouse tissue based on sequence homology. I have gone ahead and placed the order for receiving ab1659 as a replacement and your order number is*******. |

LOT NUMBER ****** ORDER NUMBER Order# ******* DESCRIPTION OF THE PROBLEM Non-specific staining of most tissues on TMA slide. Isotype control at highest dilution showed staining similar to experimental slides. SAMPLE Mouse TMA paraffin slide (liver, spleen, intestine, tumor, etc...) PRIMARY ANTIBODY 60 minute incubation up to 15ug/ml DETECTION METHOD DAB POSITIVE AND NEGATIVE CONTROLS USED 15ug/ml non-immune rabbit IgG used as isotype control. ANTIBODY STORAGE CONDITIONS -20C undiluted FIXATION OF SAMPLE 10% NBF 24 hours ANTIGEN RETRIEVAL High pH, middle, low, unitrieve, and proteinase K. PERMEABILIZATION STEP Peroxidase block with methanol BLOCKING CONDITIONS Only peroxidase block. SECONDARY ANTIBODY Envision Rabbit Dako HOW MANY TIMES HAVE YOU TRIED THE APPLICATION? 17 HAVE YOU RUN A "NO PRIMARY" CONTROL? Yes DO YOU OBTAIN THE SAME RESULTS EVERY TIME? No WHAT STEPS HAVE YOU ALTERED? No staining for any retrievals until the concentration was increased from the recommended .5-1ug/ml up to 15ug/ml. Then the staining was non-specific and present in the isotype controls. ADDITIONAL NOTES We really need CCR6 to be stained in FFPE mouse tissue. I would like to try another CCR6 (ab1659). It is listed on LSBio as working in mouse as well as human. Could you replace the ab78429 that is not working with ab1659? |

ANSWER: |
I have looked over all the information that you sent and you have tried all the variations in protocol that I can think of to try and get ab78429 to work in IHC. I would be happy to send you a replacement antibody however I would just like to say that we do not guarantee ab1659 to work in mouse tissue. The data we have on it suggests that at best it will react weakly but it does not fall under our Abpromise. If you would still like to try the antibody I will be more than happy to send you a vial but I just wanted to let you know that we do not guarantee it to work in mouse tissue. |
