Recommended dilutions
WB: 1/1000. Predicted molecular weight: 85 kDa.
IHC-P: Use at an assay dependent dilution.
IHC-Fr: Use at an assay dependent dilution.
For an ABC system, dilute 1:25-50 and incubate for 30-60 minutes at RT.
For formalin fixed paraffin embedded sections, perform heat mediated antigen retrieval before commencing with IHC.
With high pH retrieval solutions, some tissues may require an avidin-biotin block.
Overstaining with hematoxylin can mask staining, thus reducing overall contrast and making interpretation difficult.
It is therefore recommended that a light hematoxylin stain be applied.
Recently, CD10 has been used in a panel for mantle cell lymphoma with cyclin D1(+), CD43 (+), CD5 (+), IgM (+), CD23 (-) and CD10 (-).

Relevance 

CD10, also known as the Common Acute Lymphocytic Leukemia Antigen (CALLA), is a cell surface enzyme with neutral metalloendopeptidase activity which inactivates a variety of biologically active peptides. CD10 is expressed on the cells of lymphoblastic, Burkitt's, and follicular germinal center lymphomas, immature B cells with in adult bone marrow and on cells from patients with chronic myelocytic leukemia (CML). CD10 is also present on breast myoepithelial cells, bile canaliculi, fibroblasts, with especially high expression on the brush border of kidney and gut epithelial cells.
