Our Abpromise guarantee covers the use of ab79423 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
WB: 1/5000 - 1/10000.Detects a band of approximately 100 kDa (predicted molecular weight: 86 kDa).
Flow Cyt: 1/100 - 1/150.
Is unsuitable for or IP.
Thermolysin-like specificity, but is almost confined on acting on polypeptides of up to 30 amino acids. Biologically important in the destruction of opioid peptides such as Met- and Leu-enkephalins by cleavage of a Gly-Phe bond. Able to cleave angiotensin-1, angiotensin-2 and angiotensin 1-9. Involved in the degradation of atrial natriuretic factor (ANF). Displays UV-inducible elastase activity toward skin preelastic and elastic fibers.
Belongs to the peptidase M13 family.
Cell membrane.

All lanes : Anti-CD10 antibody [EPR2997] (ab79423) at 1/10000 dilution
Lane 1 : Jurkat cell lysate
Lane 2 : LnCap cell lysate
Lysates/proteins at 10 µg per lane.
Secondary
HRP labelled goat anti-rabbit at 1/2000 dilution
Predicted band size : 86 kDa
Observed band size : 100 kDa (why is the actual band size different from the predicted?)

Overlay histogram showing Jurkat cells stained with ab79423 (red line). The cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab79423, 1/100 dilution) for 30 min at 22°C. The secondary antibody used was DyLight® 488 goat anti-rabbit IgG (H+L) (ab96899) at 1/500 dilution for 30 min at 22°C. Isotype control antibody (black line) was rabbit monoclonal IgG (1µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive signal in Jurkat cells fixed with methanol (5 min)/permeabilized in 0.1% PBS-Tween used under the same conditions.
