Application notes

Recommended dilutions Flow Cyt: 1/25 - 1/200 (10µl per million cells). IHC-P: 1/25 - 1/200. Use on PERIODATE LYSINE paraformaldehyde fixed paraffin embedded tissues (the antibody DOES NOT WORK on paraformaldehyde-only fixed), perform enzymatic antigen retrieval before commencing with IHC staining protocol. IHC-Fr: Use at an assay dependent dilution. IHC-FoFr: 1/100. ICC: Use at an assay dependent dilution. P: Use at an assay dependent dilution.
Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

CD11b is implicated in various adhesive interactions of monocytes, macrophages and granulocytes as well as in mediating the uptake of complement coated particles. It is identical to CR3, the receptor for the iC3b fragment of the third complement component. It probably recognizes the RGD peptide in C3b. CD11b is also a receptor for fibrinogen, factor X and ICAM1. It recognizes P1 and P2 peptides of fibrinogen gamma chain. The Mac1 CD11b antigen is present on macrophages, granulocytes, natural killer cells, blood monocytes. CD11b is expressed on 8% spleen cells, 44% bone marrow cells and less than 1% of thymocytes and is commonly used as a microglial marker in nervous tissue.

Immunohistochemistry (Frozen sections) - CD11b antibody [M1/70.15] (ab6332)
ab6332 at 1/500 staining mouse lumbar spinal cord tissue sections (30um) by IHC-Fr. The tissue was formaldehyde fixed and blocked with 3% serum, prior to incubation with the antibody for 18 hours. A biotinylated goat anti-rat polyclonal antibody was used as the secondary. This image is courtesy of an Abreview submitted by Ms Nancy Nutile-McMenemy
