Recommended dilutions IHC-Fr: Use at an assay dependent dilution (PMID 18469117). Immunoprecipitation: The monoclonal antibody and polyclonal antisera weremixed with labeled cell lysates or purified antigens for 2 hours at 4°C followed bythe addition of 40 ml purified RG7/7.6 mouse anti-rat kappa chain coupled tosepharose 2 mg/ml. After shaking for 1 1/2 hours at 4°C, samples were washedand subjected to SDS-PAGE and autoradiography with enhancing screens (1).
FACS analysis: Each labeling step was carried out for 30 min at 0-4°C in mediumcontaining 0.01 M NaN3. Cells (5 x 107/ml) were incubated with 50 µl M1 monoclonal antibody or irrelevant monoclonal antibody, R4/18.2, as control in the first step, washed, suspended in 50 µl of FITC-F (ab')2 anti-rat IgGin the second step, and washed through a layer of fetal calf serum.
Can also be used in cytotoxicity and binding assays.

CD11b is implicated in various adhesive interactions of monocytes, macrophages and granulocytes as well as in mediating the uptake of complement coated particles. It is identical to CR3, the receptor for the iC3b fragment of the third complement component. It probably recognizes the RGD peptide in C3b. CD11b is also a receptor for fibrinogen, factor X and ICAM1. It recognizes P1 and P2 peptides of fibrinogen gamma chain. The Mac1 CD11b antigen is present on macrophages, granulocytes, natural killer cells, blood monocytes. CD11b is expressed on 8% spleen cells, 44% bone marrow cells and less than 1% of thymocytes and is commonly used as a microglial marker in nervous tissue.

ab8878 staining CD11b in mouse brain tissue section by Immunohistochemistry (Frozen sections). Tissue samples were fixed with paraformaldehyde and blocked with 5% BSA for 1 hour at 250C. The sample was incubated with primary antibody (1/100) for 12 hours at 40C in PBS. A Cy3®-conjugated donkey polyclonal to rat IgG (1/500) was used as secondary antibody.
