Anti-CD3 epsilon antibody (ab49943)

 vielen Dank für ihre Tipps, der Western Blot hat jetzt funktioniert. Können sie uns auch ihr Protokoll für die Immunhistochemie schicken? Offenbar ist dieser Antikörper doch sehr eng an die Herstellerangaben gebunden, im Unterschied zu anderen, wo wir bei Western Blots und für die Immunhistochemie ein Standardprotokoll, eben das was ich ihnen schickte, verwenden, und dort klappt´s. Die Bezahlung der Rechnung veranlasse ich umgehend. Mit freundlichen Grüßen,

Vielen Dank für Ihre Rückmeldung. Ich freue mich zu hören, dass der Antikörper ein funktionierendes Signal gibt im Western blot.

Für die Immunohistology haben wir leider kein spezifisches Protokoll. Ich kann Ihnen jedoch empfehlen, sich an den folgenden Publikationen zu orientieren, welche diesen Antikörper auf gefrorenen, respektive paraffinierten, Schnitten benutzt haben.

Is there any difference between the specificity of the Rabbit Anti-CD3 Monoclonal Antibody, Unconjugated, Clone SP7 cat# ab16669 and Rabbit Anti-CD3 epsilon Polyclonal Antibody, Unconjugated cat#ab49943? I understand that the SP7 is the tissue culture supernatant and the Anti-CD3 epsilon is a purified antibody. 

I wonder if they can be used to replace each other in canine PBMC staining. 

I just received the SP7 order but couldn't find information of the protein concentration. Will every lot of this supernatant have consistent antibody titer?

I would appreciate a prompt response.

The relative specificity of each antibody is difficult to assess without a side-by-side comparison. The data for the SP7 clone and the positive reviews we have received from customers are all encouraging.

Staining of canine samples has not been assessed for ab16669. We expect reactivity based on homology of the immunogen with canine CD3 (77% identity) but this is not covered by our guarantee.

The concentration of the SP7 tissue culture supernatant is not determined. We estimate specific antibody concentration for tissue culture supernatants in general to be about 50 ug/ml. Each lot is QC'd to ensure that there will be consistency in results from one lot to the next.
