Anti-CD36 antibody [VM58] (ab8145)

The ds when we received the antibody said tested in IHC and also tested in IHC-Fr. We assumed this meant it was tested in IHC-P and IHC-Fr. We use the Ventana system. We have tried the following HIER: citrate pH 6, EDTA pH 9 and PIER: protease 2. We use the antibody on tonsil 1/25, 1/50, 1/100, 1/500 at 37C-42C for 1-2 hrs. For secondary we use a Ventana anti-mouse anti-rabbit cocktail. Even if we do not block we do not see any staining at all.

In particular I am wondering how long you fix in formalin for, as fixation is often an over-looked cause of antibodies not working properly in IHC-P.

Do you have any sort of positive control for this antibody commercially available? If you know of any other compnaies that have one please let me know.

ANSWER:

We curently do not stock, or are aware of any other company which produces, a commercially available control for this antibody. Since this antibody cross reacts with inflammatory cells and b-cells, maybe a section of lymphoid tissue such as spleen or tonsil, or any tissue with an inflammatory condition will suffice. However, since we have not tested the antibody on such tissues we can't guarantee results.

I read that for fluorescence technique on cells fixation it is advised. I cannot fix cells before the staining for the analysis by cytolfuorometry is the antibody working well on live cells.

ANSWER:

The antibody ab8145 to CD36 will work well with live cells. However, precautions should be taken to avoid aggregation of platelets that might be caused by this antibody.
