Recommended dilutions IHC-P: Use at an assay dependent concentration. IHC-Fr: Use at an assay dependent concentration. This antibody may be diluted to a titer of 1:25-1:50 in an ABC method. We suggest an incubation period of 1-2 hours at room temperature. However, depending upon the fixation conditions and the staining system employed, optimal incubation and dilutions should be determined by the user. Heat mediated antigen retrieval of formalin fixed paraffin embedded tissues may be required before immunostaining. Use peroxidase block after the primary antibody or staining will be diminished. Post-fixation with 10% formalin after the primary antibody may improve antibody staining.
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Relevance
|
CD4 is a single chain transmembraneous glycoprotein (59 kDa) which belongs to the immunoglobulin superfamily. CD4 is present on a subset of T lymphocytes ("helper/inducer" T cells) and is also expressed at a lower level on monocytes, tissue macrophages and granulocytes. The antigen is involved in binding to MHC class II molecules. The intracellular domain of the antigen is associated with p56lck protein tyrosine kinase.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - CD4 antibody [BC/1F6] (ab846)
ab846 at 1/250 staining human PBMC tissue sections by IHC-P. The tissue was paraformaldehyde fixed and then an antigen retieval step was performed (heat mediated) prior to incubating the cells with the antibody for 20 minutes. A biotinylated goat anti-mouse IgG antibody was used as the secondary. This image is courtesy of an Abreview submitted by Miss Jill Montgomery

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - CD4 antibody [BC/1F6] (ab846)
ab846 staining CD4 in Human inflammatory dermatosis tissue sections by IHC-P (formaldehyde fixed paraffin embedded sections). Tissue was fixed with formaldehyde and blocked by 2% BSA for 30 minutes at 22°C. Antigen retrieval was by heat mediation in citrate buffer (pH 6.0). Samples were incubated with primary antibody (1/25 in BSA/TBS) for 14 hours at 4°C. Ab 6785 (1/800) was used as the secondary antibody This image is courtesy of an Abreview submitted by Dr. Sandra Peternel
