Flow Cyt Use 10µl for 106 cells. CD4 immunofluorescence analysis can be performed on a flow cytometerequipped with an excitation source of 488nm and fitted with logarithmic amplifiers.
ICC/IF Use at an assay dependent dilution.
1. Counting and identification of the T-helper/inducer lymphocyte subset in human lysed whole peripheral blood or mononuclear cells separated by density gradient. 2. Studies of HIV infection. Patients infected with HIV lose CD4+ and contemporarilyincrease CD8+ lymphocyte subset 3. Characterization of subtypes of T-cell leukaemias and lymphomas.

  • FunctionAccessory protein for MHC class-II antigen/T-cell receptor interaction. May regulate T-cell activation. Induces the aggregation of lipid rafts.

    modifications
    Palmitoylation and association with LCK contribute to the enrichment of CD4 in lipid rafts.
  • Cellular localizationCell membrane. Localizes to lipid rafts. Removed from plasma membrane by HIV-1 Nef protein that increases clathrin-dependent endocytosis of this antigen to target it to lysosomal degradation. Cell surface expression is also down-modulated by HIV-1 Envelope polyprotein gp160 that interacts with, and sequesters CD4 in the endoplasmic reticulum.
