I am a researcher who is using anti-CD40 (ab58612) in immunocytochemical and immunohistochemical analyses.
I am using a CD40 knock-out mouse (cd40-/-) in which exon 3 of the cd40 gene has been targeted for disruption by a neomycin cassette (ref: Kawabe et al, 1994: http://www.sciencedirect.com/science/article/pii/1074761394900957).
I understand that the sequence of your immunising peptide is proprietary, however could you please inform me if this antibody should detect cd40 in these cd40-/- mice, i.e. does it bind any part of the protein that is encoded by exon 3 or downstream of exon 3?
This information is critical to validating this antibody for our use. I respect you cannot inform me of the sequence that the antibody is raised against, but if you
could tell me whether this antibody will detect cd40 in mouse cd40-/- tissues, I would be most grateful.
Additionally, If you could recommend an antibody that would be useful to detect CD40 in wild type tissues but should be negative in cd40-/- mice, this would be most helpful.
