  • Applications

Our Abpromise guarantee covers the use of ab83917 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

    Application Notes
SDS-PAGE SDS-PAGE: Use at an assay dependent dilution. pI: ab83917 separates into a number of glycoforms with an observed pI between 5.5 and 9.0 in 2D PAGE due to post-translational modifications, in particular glycosylation. This compares with the unmodified CD40- Fc Chimera that has a predicted pI of 6.0.
WB WB: Use at an assay dependent dilution. Ab83917 can be used as a WB positive control in conjunction with ab13545. Molecular Mass: Under reducing conditions ab83917 migrates as a broad band between 50 and 65 kDa on SDS-PAGE due to post-translational modifications, in particular glycosylation. This compares with unmodified CD40-Fc Chimera that has a predicted monomeric molecular mass of 46.5 kDa.
Functional Studies FuncS: Use at an assay dependent dilution.

  • FunctionReceptor for TNFSF5/CD40LG.
  • Tissue specificityB-cells and in primary carcinomas.
  • Involvement in diseaseDefects in CD40 are the cause of hyper-IgM immunodeficiency syndrome type 3 (HIGM3) [MIM:606843]; also known as hyper-IgM syndrome 3. HIGM3 is an autosomal recessive disorder which includes an inability of B cells to undergo isotype switching, one of the final differentiation steps in the humoral immune system, an inability to mount an antibody-specific immune response, and a lack of germinal center formation.

    • Densitometry of protein isoforms visualised by 2-DE.
The densitometry scan demonstrates the purified human cell expressed protein exists in multiple isoforms, which differ according to their level of post-translational modification.
The triangle indicates the theoretical MW and pI of the protein.
