Question 1

Tuesday 17-April-2012

I am looking at your ELISA detection kit for CD44(v6) - product #AB119519. I am looking into an ELISA based method of examining changes in phosphorylation of v6 in a proprietary system, and though your kit might be a good start. My question is this - the product page states the the kit is 1x96 wells, but page 21 of the protocol only shows 32 wells worth of testing. Are all wells in a 96-well plate coated with anti-CD44v6 and you just use as many as you need? Or are there only 32 wells available. Also, is the control the full v6 protein or merely the v6-specific peptide? If it is the full protein, is it available for separate sale? Also, for product #AB63389, do you have any data of this antibody being used against CD44 variant isoforms? By sequence they should all be reactive, but I was hoping you might have more information. Thanks so much.

ANSWER:

Thank you for contacting us.

After reading the protocol I am sure that the whole plate is coated with the antibody as all the reagents are designed to be sufficient for 12 stripes. As mentioned in the protocol, the 32 wells are just an example. Unfortunately, we do not offer the protein separated for sale, and in addition I have contacted the lab to check if this is the whole protein or just a fragment.

Regarding your second question, we have not specificly tested the antibody if it will react with all the isoforms, but you are correct, it should react with most forms except for isoform 2,9,15 out of the 17 human isoforms based on the sequence.

I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.
