  • Product overview

ab119519 CD44var (v6) Human ELISA kit (with color giving dyes) is an enzyme-linked immunosorbent assay for the quantitative detection of human sCD44var (v6). ab119519 is for research use only. Not for diagnostic or therapeutic procedures.

Alternative splicing of CD44 mRNA gives rise to the different isoforms, containing inserts of varying sizes in the extracellular domain of the molecule (exons v2-v10). All CD44 isoforms are variably glycosylated. In contrast to standard CD44 (CD44std) which is almost ubiquitously expressed, the variety of CD44 isoforms (CD44var) have a much more restricted distribution, e.g., on activated lymphocytes and macrophages (exon v6).

ab119519 detects all circulating CD44 isoforms comprising the sCD44var (v6) sequences.

Determination of sCD44var (v6) will provide more detailed insight into different pathological modifications during cancer and other diseases.

In order to help avoid any mistakes in pipetting, our kit provides color giving dyes that help monitor the addition of a solution to the reaction. Each dye gives a distinctive color for each step of the ELISA procedure.

Properties

  • FunctionReceptor for hyaluronic acid (HA). Mediates cell-cell and cell-matrix interactions through its affinity for HA, and possibly also through its affinity for other ligands such as osteopontin, collagens, and matrix metalloproteinases (MMPs). Adhesion with HA plays an important role in cell migration, tumor growth and progression. Also involved in lymphocyte activation, recirculation and homing, and in hematopoiesis. Altered expression or dysfunction causes numerous pathogenic phenotypes. Great protein heterogeneity due to numerous alternative splicing and post-translational modification events.
  • Tissue specificityIsoform 10 (epithelial isoform) is expressed by cells of epithelium and highly expressed by carcinomas. Expression is repressed in neuroblastoma cells.
  • Sequence similaritiesContains 1 Link domain.
  • DomainThe lectin-like LINK domain is responsible for hyaluronan binding.
  • Post-translational
    modifications
Proteolytically cleaved in the extracellular matrix by specific proteinases (possibly MMPs) in several cell lines and tumors.
    N-glycosylated.
O-glycosylated; contains more-or-less-sulfated chondroitin sulfate glycans, whose number may affect the accessibility of specific proteinases to their cleavage site(s).
Phosphorylated; activation of PKC results in the dephosphorylation of Ser-706 (constitutive phosphorylation site), and the phosphorylation of Ser-672.
  • Cellular localizationMembrane.

After reading the protocol I am sure that the whole plate is coated with the antibody as all the reagents are designed to be sufficient for 12 stripes. As mentioned in the protocol, the 32 wells are just an example....
