Question 1

Thursday 22-March-2012

Thanks for your answer. What about time and temperature of incubation? In which conditions did you performed the staining? Thank you very much.

ANSWER:

Thank you for contacting us.

Our labs follow the suggested protocols posted within our Protocol and Troubleshooting Page athttp://www.abcam.com/index.html?pageconfig=popular_protocols. The indirect flow cytometry protocol may be accessed directly at this page:http://www.abcam.com/index.html?pageconfig=resource&rid=11381. Within this protocol you will find that our recommended time and temperature for incubation of these antibodies is ˜30m minutes at room temperature. Please be aware that differences in cell type will require optimization of this protocol experimentally.

I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.

Question 2

Wednesday 21-March-2012

Hello, I have just received the product anti-CD45RC antibody (MRC OX-22) (Biotin) (ab33937). It would be pleasure if you can tell me in which condition you developed the flow cytrometric experiment which result you publish in the datasheet. I am interested in knowing the concentration of fluorochrome conjugated streptoavidin that you used. Thanks for your help.

ANSWER:

Thank you for contacting us.

Optimal dilution for AB33937 from our QC testing is 1:2. We used 10ul of this working dilution to stain one million cells in 100ul total volume (an additional 1:10 dilution). For the streptavidin detection reagent, we used STAR4ASTREPTAVIDIN: RPE which is a PE conjugate. We used this undiluted (again with the 1:10 dilution which is part of our flow cytometry protocol).

I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.
