Recommended dilutions Flow Cyt: Use at an assay dependent concentration. FuncS: Use at an assay dependent dilution. ICC/IF: Use at an assay dependent dilution PubMed: 17911601. Flow Cyt: Use at an assay dependant concentration. FuncS: Use at an assay dependant dilution. MEM-43 inhibits the complement-protecting activity of CD59. IP: Use at an assay dependant concentration. WB: Use at an assay dependant concentration. Use under non-reducing conditions. IHC-P: Use at an assay dependent dilution. Perform heat mediated antigen retrieval before commencing with IHC staining protocol. Sodium citrate buffer pH6.0 is recommended for this purpose. IHC-Fr: Use at an assay dependent dilution.
Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Relevance
|
CD59, an LY6 like protein expressed in human lymphoid cells, regulates the action of the complement membrane attack complex on homologous cells. It is a potent inhibitor of the complement membrane attack complex action. It acts by binding to the C8 and/or C9 complements of the membrane attack complex, thereby preventing incorporation of the multiple copies of C9 required for complete formation of the osmolytic pore. This inhibitor appears to be species-specific. CD59 is also involved in signal transduction for T-cell activation complexed to a protein tyrosine kinase.

Notes
|
When originally tested in WB, SDS was included in the sample buffer, however, feedback from one researcher has shown that it is best to omit SDS from the sample buffer.

Flow Cytometry - CD59 antibody [MEM-43] (ab9182)
ab9182 staining CD59 in Human fibrosarcoma (HT1080) by Flow Cytometry. Cells were fixed with paraformaldehyde. The sample was incubated with the primary antibody,10 µg/ml in PBS, for 1 hour at 20°C. An Abcam PE-conjugated donkey polyclonal to mouse IgG ( ab7003), 5 µg/ml, was used as secondary antibody. This image is courtesy of an anonymous Abreview
