This antibody is specific to a 53 kD protein, expressed on activated platelets. CD63 is a lysosomal membrane glycoprotein that is translocated to plasma membrane after platelet activation. It is also expressed on granulocytes, B cells and T cells.

Recommended dilutions IHC-P: Use at an assay dependent concentration. IHC-Fr: Use at an assay dependent concentration.
This antibody may be diluted to a titer of 1:100 to 1:200 in an ABC method. However, depending on the staining conditions employed, we suggest that final dilution should be determined by the user. High temperature antigen unmasking with 10mM citrate buffer, pH 6.0 is required prior to immunostaining. We suggest an incubation period of 30 minutes at room temperature. Not tested in other applications.
Optimal dilutions / concentrations should be determined by the end user.

Relevance

CD63 (Melanoma 1 antigen) is a glycoprotein of 40-60 kDa belonging to tetraspans family (TM4SF). CD63 is intracellular lysosomal, endosomal and granule protein that is translocated to cell surface upon activation of platelets, endothelial cells, and granulocytes (activation marker). CD63 is also expressed on monocytes, macrophages and endothelium. The cellular function has not been exactly elucidated. CD63 is thought to be associated with the early stages of melanoma tumor progression (in regulation of motility and adhesion of melanoma cells).
CD63 is expressed on activated platelets. CD63 is a lysosomal membrane glycoprotein that is translocated to plasma membrane after platelet activation. It is also present in monocytes and macrophages and is weakly expressed in granulocytes, B, and T cells. CD63 is identical to the melanoma-associated antigen ME491 and to the platelet antigen PTLGP40.
