Recommended dilutions
WB: Use at a concentration of 1 µg/ml. Detects a band of approximately 17 kDa (predicted molecular weight: 16 kDa). Can be blocked with CENPA peptide - di methyl K9 (ab13488).
We have had customer reports that this antibody does not appear to be suitable for ICC/IF in either HeLa, SK-N-SH or 10T1/2 cells. ab4488 should only stain at the centromeres, however in colocalization studies with ACA (anti-centromeric antigen), ab4488 was distributed throughout the nucleus and did not colocalised with ACA.
Not yet tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.

Centromeres are the differentiated chromosomal domains that specify the mitotic behavior of chromosomes. CENPA encodes a centromere protein which contains a histone H3 related histone fold domain that is required for targeting to the centromere. CENPA is proposed to be a component of a modified nucleosome or nucleosome-like structure, in which it replaces 1 or both copies of conventional histone H3 in the (H3-H4)2 tetrameric core of the nucleosome particle.
