Ueberham et al (Neuroscience, 2003)report "In human hippocampal protein extracts two distinct bands are detectable with a more intensive reactivity at about 37-38 kDa, representing the CTGF protein, and at about 42-44 kDa likely representing a glycosilated ... or a phosphorylated form of CTGF...".

Recommended dilutions IP: Use at an assay dependent dilution. PubMed 16100286. IHC-F: 1/1500 (Tissue postfixed for 4 days at 4C in 4% paraformaldehyde in 0.1M PB, then immersed in 15% sucrose for 24hrs, 30% sucrose for 48hrs and sections were cut on a freezing microtome). Use Biotinylated secondary antibody then stextra-avidin-peroxidase chromogenic detection. (See Ueberham et al for details). IHC-P: 1/400 - 1/800. (See Razzaque et al and Candido et al for details). Antigen retrieval can be avoided. IHC-Fr: 1/500 (PMID 19299462). WB: 1/5000. Can be blocked with CTGF (C-term) peptide (mouse). Block membrane in 4% BSA in Tris-buffered saline containing 0.05% Tween 20 (TBST).(see Fu M et al.).
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Relevance
|
CTGF (connective tissue growth factor) is a 38kDa, cysteine-rich, secreted peptide. It is a new member of the peptide family that include serum-induced immediate early gene products, a v-src-induced peptide and a putative proto-oncogene. Among the many functions of the CTGF gene family are embryogenesis, wound healing and regulation of extracellular matrix production.

Immunohistochemistry (Paraffin-embedded sections) - CTGF antibody (ab6992)
ab6992 at a 1/100 dilution staining CTGF from Mouse Tissue sections (Skin) by Immunohistochemistry (Formalin-fixed paraffin-embedded sections). The antibody was incubated with the tissue for 12 hours and then detected using a Labeled AP polymer. The right side of image is regrown wounded epidermis and shows significant staining for CTGF. This image is courtesy of an Abreview submitted by Martina Ziegenbein on 6 January 2006.
