Recommended dilutions ELISA:To detect mCTLA-4 by direct ELISA (using 100µl/well antibody solution) a concentration of at least 0.5µg/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with compatible secondary reagents, allows the detection of 0.2 - 0.4 ng/well of recombinant mCTLA-4.
Western Blot:To detect mCTLA-4 by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 µg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant mCTLA-4 is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.

Relevance
|
CTLA4 has a critical role in T cell differentiation and contributes to the maintenance of peripheral T cell tolerance to self antigens. It exerts its major effects on T cell immune responses via regulation of the cell cycle and may be involved in T cell activation. It binds to B7-1 (CD80) and B7-2 (CD86), and it contains one immunoglobulin-like V type domain. It is widely expressed, with highest levels in lymphoid tissues.
