    Abcam's Human CXCL7 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of human CXCL7 in serum, plasma and cell culture supernatants.

     

    This assay employs an antibody specific for human CXCL7 coated on a 96-well plate. Standards and samples are pipetted into the wells and CXCL7 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated antihuman CXCL7 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CXCL7 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

    CXCL7 which is a protein with a length of 70 amino acids, belongs to the family of chemotactic cytokines. The expression of CXCL7 is induced in human monocytes by treatment with bacterial endotoxins. It stimulates the release of elastase from neutrophils and shows some of the activities of IL8. CXCL7 interacts with the NAP-1 receptor and binds to one type of receptors that also binds IL8 and related factors.

  • RelevanceCXCL7 is a platelet-derived growth factor that belongs to the CXC chemokine family. It is a potent chemoattractant and activator of neutrophils and has been shown to stimulate various cellular processes including DNA synthesis, mitosis, glycolysis, intracellular cAMP accumulation, prostaglandin E2 secretion, and synthesis of hyaluronic acid and sulfated glycosaminoglycan. It also stimulates the formation and secretion of plasminogen activator by synovial cells.

      The Assay Diluent B (1X) should be used to dilute the antibodies, regardless of what kind of samples are being assayed, plasma or other. Remember to dilute the diluent 5-fold (step 3): one part diluent stock and 4 parts deionized or distilled water.

      I ordered the ab100613 CXCL7 Human ELISA kit from Abcam. Can I check what is the format of the 96 well plate for this ELISA product? Is the format according to the Corning, Nunclon, BD Falcon, or Greiner 96 well plate?
