Relevance 

Voltage-sensitive calcium channels (VSCC) mediate the entry of calcium ions into excitable cells and are also involved in a variety of calcium-dependent processes, including muscle contraction, hormone or neurotransmitter release, gene expression, cell motility, cell division and cell death. The isoform alpha-1D gives rise to L-type calcium currents. Long-lasting (L-type) calcium channels belong to the "high-voltage activated" (HVA) group. They are blocked by dihydropyridines (DHP), phenylalkylamines, benzothiazepines, and by omega-agatoxin-IIIA (omega-Aga-IIIA). They are however insensitive to omega-conotoxin-GVIA (omega-CTx-GVIA) and omega-agatoxin-IVA (omega-Aga-IVA).


Alpha 1-subunits of voltage-gated Ca2+ channels are highly sensitive to proteases. All procedures involving a full-length protein should be performed at 4°C and include the following protease inhibitor cocktail: pepstatin A (1 µg/ml), leupeptin (1 µg/ml), aprotinin (1 µg/ml), benzamidine (0.1 µg/ml), AEBSF (0.2 mM) and calpain inhibitors I (LLNL, 8 µg/ml) and II (ALLM, 8 µg/ml).

To reduce nonspecific staining, centrifuge all antibody preparations before use (10000 x g for 5 min) to remove any aggregates. Working dilutions of antibody should be made in a physiological buffer containing a carrier protein such as 1-3% BSA.

ab54421 is supplied with a control peptide (20 µg, lyophilized from PBS). Reconstitute the peptide with 100 µl dH2O, aliquot, and freeze (-20°C). Antibody binding can be fully inhibited with the control blocking peptide. For use as a negative control, preincubate 1 µg control peptide with 1 µg antibody for 1 hr at room temperature.
