Specificity

ab54421 recognizes all forms of the Alpha 1D-subunit of the voltage-gated calcium channel. The epitope is specific for Alpha 1D-subunits of the voltage-gated Ca2+ channel and is not present in any other known protein.

Primary antibody notes

Alpha 1-subunits of voltage-gated Ca2+ channels are highly sensitive to proteases. All procedures involving a full-length protein should be performed at 4°C and include the following protease inhibitor cocktail: pepstatin A (1 µg/ml), leupeptin (1 µg/ml), aprotinin (1 µg/ml), benzamidine (0.1 µg/ml), AEBSF (0.2 mM) and calpain inhibitors I (LLNL, 8 µg/ml) and II (ALLM, 8 µg/ml). To reduce nonspecific staining, centrifuge all antibody preparations before use (10000 x g for 5 min) to remove any aggregates. Working dilutions of antibody should be made in a physiological buffer containing a carrier protein such as 1-3% BSA. ab54421 is supplied with a control peptide (20 µg, lyophilized from PBS). Reconstitute the peptide with 100 µl dH2O, aliquot, and freeze (-20°C). Antibody binding can be fully inhibited with the control blocking peptide. For use as a negative control, preincubate 1 µg control peptide with 1 µg antibody for 1 hr at room temperature.

Function

Voltage-sensitive calcium channels (VSCC) mediate the entry of calcium ions into excitable cells and are also involved in a variety of calcium-dependent processes, including muscle contraction, hormone or neurotransmitter release, gene expression, cell motility, cell division and cell death. The isoform alpha-1D gives rise to L-type calcium currents. Long-lasting (L-type) calcium channels belong to the 'high-voltage activated' (HVA) group. They are blocked by dihydropyridines (DHP), phenylalkylamines, benzothiazepines, and by omega-agatoxin-IIIA (omega-Aga-IIIA). They are however insensitive to omega-conotoxin-GVIA (omega-CTx-GVIA) and omega-agatoxin-IVA (omega-Aga-IVA).

Tissue specificity

Expressed in pancreatic islets and in brain, where it has been seen in cerebral cortex, hippocampus, basal ganglia, habenula and thalamus. Expressed in the small cell lung carcinoma cell line SCC-9. No expression in skeletal muscle.

Domain

Each of the four internal repeats contains five hydrophobic transmembrane segments (S1, S2, S3, S5, S6) and one positively charged transmembrane segment (S4). S4 segments probably represent the voltage-sensor and are characterized by a series of positively charged amino acids at every third position.
