ab11426 Calbindin antibody - Neuronal Marker Can you clarify how the specificity of this product was determined for each tested application? I need this information in order to publish my manuscript. |
|||||||||
ANSWER: |
Thank you for your patience in this matter. The originator of ab11426 (Calbindin antibody - Neuronal Marker) has provided a reference, which describes the characterization of this product in immunoprecipitation, Western blotting, ELISA and immunohistochemistry. The reference is: N. Kurobe et. al. Development and Age-Dependent Changes of 28-kDa Calbindin-D in the Central Nervous Tissue Determined with a Sensitive Immunoassay Method. J. Neurochem. 58: 128-134, 1992 I hope this helps. Please do not hesitate to contact me should you have further questions. |

Western blot of calbindin D from rat kidney using ab11426.
ab11426 at a dilution of 1/1000, staining Calbindin in neurons (Alexa 488 secondary at 1/2000) on 30
NB: No labeling observed following omission of primary antibody.
Sections were viewed using an Axioplan 2 Imaging microscope (Imaging Associates) fitted with 10x, 20x and 40x Plan-Neofluorobjectives (Zeiss, Germany) and images were taken using a AxioCam Hrm digital camera (Zeiss, Germany) and AxioVision software (Imaging Associates). Imgae recoloured in Photoshop.
Mouse Embryonic Stem Cells immunopositive for Calbindin (Abcam ab11426, 1:500). Secondary antibody: Alexa Fluor 488 goat anti-rabbit IgG (1:500). Cells underwent a 5-stage differentiation protocol to generate neurons. These cells were stained after stage 5.
